A novel experimental compound called PA-457 is set to begin human clinical trials this month, according to New Scientist. The drug belongs to a newly identified category known as maturation inhibitors, the first of its kind designed to address HIV strains that have grown resistant to standard antiretroviral medications.
PA-457 works by disrupting the capsid protein, a cone-shaped structure that shelters the genetic core of HIV particles as they emerge from infected immune cells. When that structure is compromised, the virus loses its ability to infect additional cells.
The upcoming trial will enroll 48 HIV-positive patients whose current drug regimens are no longer effective. Half will receive PA-457 alongside their existing medications, while the other half will receive a placebo. Earlier laboratory and small-scale human tests showed the drug reduced HIV levels in the bloodstream tenfold within hours.
The new phase will examine how PA-457 performs in combination with other HIV treatments, rather than alone. Extensive additional trials will still be required to confirm its safety and effectiveness, meaning the drug is likely years away from broad availability. Panacos Pharmaceuticals, based in Gaithersburg, Maryland, is leading its development.
